ABSTRACT

Objective • To analyze the differentially expressed profiles of long noncoding RNA (lncRNA) in endometrial cancer (EC) tissues and normal endometrial tissues. Methods • The RNA was extracted from 21 EC tissues and 5 normal endometrial tissues, respectively, and lncRNAs expression profiles were analyzed and screened by transcriptome sequencing technology. Gene Ontology (GO) function analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were carried out for the differentially expressed lncRNAs, and their expression differences between the transcriptome sequencing and TCGA database were analyzed. Results • There were 3 060 differentially expressed lncRNAs, of which 2 046 were upregulated and 1 014 were down-regulated. GO functional analysis showed that these lncRNAs were associated with cell adhesion, immune response, inflammatory response and cell proliferation. KEGG pathway analysis showed that these lncRNAs were mainly enriched on the pathways, such as PI3KAkt signaling pathway, cell adhesion and cytokine-cytokine receptor interaction. Intersection analysis showed that 57 lncRNAs were up-regulated or downregulated simultaneously in the sequencing results and TCGA database. Conclusion • The expression of lncRNAs in EC tissues and normal endometrial tissues are significantly different, suggesting that it may play an important role in the occurrence and development of EC.

ABSTRACT

Objective·To analyze the clinical and pathological characteristics of endometrial carcinoma patients before and after menopause. Methods·Fifty-two premenopausal cases and 150 postmenopausal cases with endometrial carcinoma from December 2013 to March 2017 in Ruijin Hospital, Shanghai Jiao Tong University School of Medicine were retrospectively analyzed. Results·The ratio of patients with hypertension in premenopausal group was 28.8%, which was significantly lower than that in postmenopausal group (P<0.01). In premenopausal group, the proportion of G3 in histological grading was 8.3%, and the ratio of deep myometrial invasion was 13.5%, all of which were significantly lower than those of postmenopausal group (P<0.05). The ratio of ovarian involvement was as high as 17.6%, which was significantly higher than that of postmenopausal group (P<0.05). There were no significant differences between the two groups in other clinical-pathological parameters such as obesity, diabetes, CA125, CA199, surgical-pathological staging, pathological type, vascular involvement, and lymph node metastasis. Conclusion·Premenopausal endometrial carcinoma is less malignant, while the probability of ovarian involvement is higher.

ABSTRACT

<p><b>BACKGROUND</b>Two recent whole-exome sequencing researches identifying somatic mutations in the ubiquitin-specific protease 8 (USP8) gene in pituitary corticotroph adenomas provide exciting advances in this field. These mutations drive increased epidermal growth factor receptor (EGFR) signaling and promote adrenocorticotropic hormone (ACTH) production. This study was to investigate whether the inhibition of USP8 activity could be a strategy for the treatment of Cushing's disease (CD).</p><p><b>METHODS</b>The anticancer effect of USP8 inhibitor was determined by testing cell viability, colony formation, apoptosis, and ACTH secretion. The immunoblotting and quantitative reverse transcription polymerase chain reaction were conducted to explore the signaling pathway by USP8 inhibition.</p><p><b>RESULTS</b>Inhibition of USP8-induced degradation of receptor tyrosine kinases including EGFR, EGFR-2 (ERBB2), and Met leading to a suppression of AtT20 cell growth and ACTH secretion. Moreover, treatment with USP8 inhibitor markedly induced AtT20 cells apoptosis.</p><p><b>CONCLUSIONS</b>Inhibition of USP8 activity could be an effective strategy for CD. It might provide a novel pharmacological approach for the treatment of CD.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the differences of psychological and behavioral development between children aged 1 to 3 years fostered by grandparents and those by parents.</p><p><b>METHODS</b>Psychological and behavioral development of 443 children aged 1 to 3 years fostered by their grandparents and of aged-matched 443 children fostered by their parents were assessed with DST, an intellectual developmental screening test developed by Pediatric Hospital of Fudan University in Shanghai.</p><p><b>RESULTS</b>The abilities of social adaptation and intelligence development in children fostered by their grandparents were obviously retarded as compared with those in children fostered by their parents.</p><p><b>CONCLUSIONS</b>There are shortcomings in psychological and behavioral development in children aged 1 to 3 years fostered by grandparents.</p>
